Detection of protein on the strip after IEF

Postby Wan Atiqah » Wed Sep 05, 2012 1:55 am

Hi there,
I would like to ask how can i detect the protein is existed on the strip after IEF? I am facing problem that the spot did'nt expressed on the gel after staining it.

1.Can I stain the strip with Coomassie stain after IEF?
2. If yes, how it would affect the protein expression on the gel afterwards?

Thank you.. :)

1. Yes, you can Coomassie stain an IEF strip
2. If you are planning on continuing on to SDS-PAGE, I would recommend that you use a Coomassie stain that doesn't fix the protein in the IEF strip or maybe try using a Zinc stain, which has a reversible fix. If you do use a fix you will likely find that a portion of your sample stays in the IEF strip.
